A Breathrough in Cryo-EM and Cryo-Tomography:

Identification of nuclear proteins and their structure in live cells

Recently it was reported that our Triple Gold Halo (3-in-1 Halo-Alexa®-Nanogold®) penetrated live cells, entered the nucleus, and specifically bound Halo-tagged histone variant H2AFY.

Live cells were quick frozen and the histone in situ structure elucidated by cryo-tomography.

The small 1.4 nm Nanogold® probe gets into the nucleus, and was used to identify histones and their structures in “live” cells.

3-in-1 Triple Gold Halo tags carry a tiny, 1.4 nm Nanogold® probe, used to penetrate the nucleus and identify histones and their structure in live cells!

“…No [other] exogenous, e.g., antibodies or endogenous, e.g., small genetically encoded proteins visible in cryo-EM, have been able to specifically target and label histone-specific nucleosomes with minimal disruptions to the chromatin landscape.”

“1.4-nm Halo-Alexa-Nanogold-594, 1.4-nm Halo-Alexa-Nanogold-488, and 5-nm Halo-Nanogold conjugates were custom ordered from Nanoprobes.”
